Harlem was founded in the 1920s during construction of the Seaboard Air Line Railway through Hendry County. The town was named after the Harlem neighborhood in New York City.

Early settlers were attracted to Harlem for the rich soil, establishing farms growing vegetables, sugar cane, and citrus. Agricultural activities fueled the town's early economy and population growth.

Today, Harlem still retains its small town rural heritage while serving as a modest bedroom community for the Central Florida area.
